Southern Terminus:  At I-90 in West Seneca

Northern Terminus:  At the Route 16 junction 1

Length:  16.9 miles

Counties: Erie

Routing: Route 16 is in effect a freeway bypass of Route 16.  Route 400 follows Route 16 to the north of West Seneca and to the east of East Aurora before the two highways converge and are co-designated along the same routing until Route 400 ends at Olean Road in South Whales.

Erie County Southbound advanced signage for the US-20A/NY-78 interchange.  This is a north-side partial diamond interchange.  South of this interchange Route 400 meets Route 16 at south-side diamond interchange.  Route 16 is multiplexed with Route 400 until South Whales.  The divided highway ends in South Whales, as does the 400 designation, though the highway continues southerly as Route 16.
